Placental Expression of Organic Anion Transporting Polypeptide -A and -C in Intrahepatic Cholestasis of Pregnancy

Objective

To investigate placental expression of cholate transporter organic anion transporting polypeptide (OATP)-A and -C during normal pregnancy and patient with intrahepatic cholestasis of pregnancy (ICP).

Methods

From May 2006 to February 2007, placentas were collected from 20 pregnancy complicated with intrahepatic cholestasis (ICP group). Meanwhile, 20 women termination pregnancy as well as plan centas from 8 1st trimester and 7 2nd trimester pregnancy were selected into control group (n=35). RT-PCR was applied for the measurement of placental OATP-A mRNA, OATP-C mRNA expression. This localization of OATP-A was analyzed by immunohistochemistry. The study protocol was approved by the Ethical Review Board of Investigation in Human Being of West China Second University Hospital, Sichuan University. Informed consent was obtained from all participants.

Results

①OATP-A mRNA was detected in placentas in control group of 1st, 2 nd, 3 rd trimester and ICP group placentas, transcript of organic anion transporting polypeptide-C was not detected in these placentas. Placental OATP-A mRNA expression was higher in ICP group than that in control group (40.72±31.46 vs. 21.54±15.65, P<0.05). No significant difference was found in placental OATP-A mRNA expression among 1st, 2nd and 3rd trimester placentas in control group. ②organic anion transporting polypeptide-A expression was detected in trophocytes by immunohistochemistry.

Conclusion

OATP-A mRNA expression is up-regulated in intrahepatic cholestasis of pregnancy placenta, which may represent a compensatory (anti-cholestatic) mechanism for increased bile acids levels in intrahepatic cholestasis of pregnancy.
