The establishment and evaluation on the newborn rats model of periventricular leukomalacia

Objective

To establish the 2-day-old SD rats model of periventricular leukomalacia (PVL) model.

Methods

A total number of 84 2-day postnatal SD rats were randomly divided into 2 groups: The control group and the PVL model group. Rats in the PVL model group were subjected to the ligation of right carotid artery, and then they were kept in a box with 6% oxygen and 94% nitrogen for 4 h to establish the PVL animal model. Morphological observation, the assessment of neural behavior and the appraisal of the growth and development have been made.

Results

The pathological changes of control group was not obvious, while distinguished periventricular white matter damage was observed at 48 h after HI and the topical pathological changes had been found at 14 d after HI under light microscope and electronic microscope. The assessment of neural behavior showed that both the function of the sense and movement and the ability of the sensibility and behavior have apparently weakened in the PVL group in comparison with the control group.

Conclusion

The establishment of PVL model was successful and accorded with the pathological changes of PVL in premature infants.
