The effects of fibrin clot and cellular growth factor on the human amniotic epithelial cells

Objective

To investigate whether human amniotic epithelial cells(HAEC)could be generate on the matrix of formulated fibrin clot in vitro and the effects of epidermal growth factor(EGF), basic fibroblast growth factor(bFGF)and transforming growth factor β1(TGF-β1)on the proliferation of HAEC.

Methods

The cultivated HAECs on the formulated fibrin clot were examined by phase contrast microscopy, Giemsa staining and scanning electron microscopy. We measured apoptosis of HAECs on the formulated fibrin clot with growth factor(EGF,bFGF, TGF-β1)by TUNEL.

Results

HAECs generate on the formulated fibrin clot in vitro. HAECs exposure to EGF or bFGF was associated with a lower rate of apoptosis than control group(P<0. 05), however, HAECs exposure to TGF-β1 showed opposite effect(P<0. 05).

Conclusion

HAECs generate well on the formulated fibrin clot in vitro. HAECs exposure to EGF or bFGF was decreased apoptosis rate than control group, however, HAECs exposure to TGF-β1 was increased effect.
