细胞凋亡异常是导致肿瘤化疗耐药的重要因素之一。目前研究证实,X染色体连锁凋亡抑制蛋白(XIAP)在细胞凋亡抑制中扮演关键角色。在多数化疗耐药的妇科肿瘤细胞中,XIAP过表达,XIAP相关因子1(XAF1)则低表达或缺失。现有研究结果发现,顺铂耐药卵巢癌中,XAF1可拮抗XIAP的抗凋亡作用,逆转XIAP对细胞的保护作用。笔者拟针对XIAP、XAF1与妇科肿瘤化疗耐药关系的研究现状进行阐述,旨在为临床防治肿瘤化疗耐药提供新思路。

Abnormality in apoptosis is one of the main causes of chemotherapy resistance. Current researches have proven that X-linked inhibitor of apoptosis protein (XIAP) plays a key role in apoptosis. In most of the tumor cells with chemotherapy resistance in gynecology, XIAP is over expressed, while XIAP-associated factor-1 (XAF1) is underexpressed or deleted. Recent researches indicate that in ovarian cancer with cisplatin resistance, XAF1 has a resistance to the effect of XIAP, and will eventually reverse the protection of XIAP to cells. This article focuses on current research on the relationship among XIAP, XAF1 and chemotherapy resistance in gynecologic cancer treatment, in order to offer some fresh thoughts for prevention and treatment of chemotherapy resistance in gynecologic tumor.
