目的

探讨不同剂量、不同疗程的米非司酮(mifepristone)对减小子宫内膜异位症(EMs)病灶的疗效。

方法

建立EMs大鼠模型,将建模成功的79只大鼠随机分为高剂量组[米非司酮剂量为2.604 mg/(kg·d)]、低剂量组[米非司酮剂量为1.041 mg/(kg·d)]及对照组(1 mL花生油),治疗期内死亡7只;将3组再按不同疗程分为20 d亚组,30 d亚组,40 d亚组,观察用药20 d,30 d及40 d时异位病灶体积的减小率,从而判断治疗的有效性。

结果

高剂量组及低剂量组分别与对照组疗效比较,异位病灶体积均显著减小,差异均有统计学意义(P<0.05);而低剂量组与高剂量组的异位病灶体积减小率比较,差异无统计学意义(P=0.662),用药20 d,30 d及40 d时病灶减小率比较,差异亦无统计学意义(P=0.559,0.145,0.057)。

结论

米非司酮可抑制EMs异位病灶的生长,低剂量组与高剂量组抑制趋势无显著差异,而且延长治疗周期,亦不能增强该药对异位内膜生长的抑制作用。

Objective

To investigate the effect of different dosages and different treatment methods of mifepristone in rat's endometriosis(EMs).

Methods

A total of 79 rats were selected to establishment of models of EMs, and were randomly divided into high-dose group[dosages of 2.604 mg/(kg·d)], low-dose group[dosages of 1.041 mg/(kg·d)], control group(placebo of 1 mL peanut oil)to determine the drugs effects of the ectopic lesions reduce by medication. There were 7 rats died during treatment period. According to different courses of treatment 20 days, 30 days and 40 days, 72 rats were divided into 9 sub-groups to determine the effectiveness of treatment.

Results

After treatment of mifepristone, ectopic lesions of EMs were significantly reduced than those before treatment in low-dose group and high-dose group (P<0.05); in low-dose group and high-dose group there were significance difference than those in control group (P<0.05); however, curative effects of mifepristone between low-dose group and high-dose group (P=0.662) or different courses of treatment 20 days, 30 days and 40 days subgroups had no statistically significant difference (P=0.559, 0.145, 0.057).

Conclusions

Mifepristone could inhibit the growth of EMs ectopic lesions. There was no statistically significant difference to treatment models of EMs rats by medication dosages of 2.604 mg/(kg·d), 1.041 mg/(kg·d) or different courses of treatment by medication 20 days, 30 days and 40 days subgroups.
