Vascular Endothelial Growth Factor and Its Receptors Expression and Microvessel Density in Ovarian Malignant Neoplasm

Objective

To investigate the expression of vascular endothelial growth factor(VEGF) and its receptors (Flt-1, KDR/Flt-1) in ovarian carcinoma and its clinical significance and the relationship with tumor microvessel density (MVD).

Methods

This was a prospective observational study with institutional ethics approval and written all participants consent. From January 2000 to June 2004, a total of 48 cases of ovarian carcinoma who diagnosis in Beijing General Army Hospital recruited. Strept-avidin-birotin-peroxidase complex (SABC) immunohistochemistry method was used to determine the expression of VEGF and Flt-1, KDR/Flk-1 in 48 cases of ovarian carcinoma. Meanwhile an antibody to von willebrand factor (FⅧ) related antigen(FⅧ-RA)were also used to detect MVD.

Results

①No significantly association were between expressive levels of VEGF and Flt-1 , KDR with pathology types, differentiated grade and clinical stage of ovarian carcinoma(P>0.05); but VEGF and Flt-1 co-expression was associated with differentiated grade and clinical stage of ovarian carcinoma (P<0.05). ②VEGF and Flt-1 expression in lymph node metastasis was higher than that in no lymph node metastasis (P<0.05). KDR expression in metastasis liver tissue was higher than that in normal liver tissue(P=0.02), VEGF and KDR co-expression was associated with level metastasis (P=0.005). ③The positive expression rate of Flt-1 of patients with the ascite ≥1000 mL were significantly higher than that in patients with the ascite <1000 mL (P=0.01). ④There was also a close relationship between VEGF and KDR expression with MVD in ovarian carcinoma (P=0.03).

Conclusions

The expression of VEGF and its receptors according many ways associated with angiogenesis, metastasis and ascite in ovarian carcinoma, They might be a biomarker for the status of present in ovarian carcinoma metastasis and prognosis.
