Effects of Proanthocyanidins on the Expression of Caspase-3 and Survivin Gene in Cervical Cancer HeLa Cell

Objective

To study the effect of proanthocyanidins (PC) on the expression of Caspase-3 and Survivin gene in cervical cancer HeLa cell.

Methods

After the HeLa cells were treated with different concentration of proanthocyanidins (0, 37.5 μmol/L, 75.0 μmol/L, 150.0 μmol/L, 300.0 μmol/L and 600.0 μmol/L). HeLa cells were divided into control group (black control), 37.5 μmol/L PC group, 75.0 μmol/L PC group, 150.0 μmol/L PC group, 300.0 μmol/L PC group and 600.0 μmol/L PC group. Cells were collected for experiments after culturing for 24 h. Expression of Caspase-3 mRNA and Survivin mRNA were detected by RT-PCR. Expression of caspase-3 protein and survivin protein were detected by Western blot, and caspase-3 relative activities were determined by color metric assay.

Results

Expression of Caspase-3 mRNA and caspase-3 protein increased with proanthocyanidins concentration from 0 to 600.0 μmol/L, but those of Survivin mRNA and survivin protein decreased. In addition, the activity of caspase-3 was increased (P<0.05), and reaching the peak in 600.0 μmol/L in PC group.

Conclusion

According to increase of concentration of proanthocyanidins, expression of the Caspase-3 mRNA and its protein, and the caspase-3 relative activities in a dose-dependent manner increased. Otherwise, expression of the survivin mRNA and its protein decreased by increasing of concentration of proanthocyanidins, also in a dose-dependent manner.
