Effects of the neonatal handling and the enriched environment on nerve growth factor and its receptor Trk A in the brain of neonatal rats

Objective

To explore the effects of the neonatal handling and the enriched environment on nerve growth factor (NGF) and its tyrosine kinase receptor A (Trk A) in the forehead cortex and hippocampus of neonatal rats.

Methods

60 neonatal rats were divided into the early intervention group and the control group randomly. The rats in early intervention group received the neonatal handling for14 d and then were kept in an enriched environment for another 14 d. NGF levels were determined by an enzyme-linked immunosorbent assay (ELISA) method and positive the cells expressed Trk A were stained by immunohistochemistry.

Results

In the forehead cortex and hippocampus, NGF levels were(2343. 58±133. 15) pg/g and(1547. 42 ± 65. 32) pg/g in the intervention group, which were much higher than those in the control group [(2174. 70±165. 21) pg/g and(1475. 23 v 76. 72)pg/g] (P<0. 05). The numbers of neurons labelled with Trk A antibody in the forehead cortex and hippocampus of the intervention group were (80. 56 ± 20. 12)cells/HF and(66. 42 ± 21. 83)cells/HF, which were more than those in the control group [(63. 50±18. 23)/HF and(50. 45 ± 17. 28)/HF](P<0. 05).

Conclusion

The neonatal handling and the enriched environment may improve brain development by increasing the levels of endogenetic NGF levels and Trk A expressed in the brain.
