We investigated the expression of the hypoxia-inducible transcription factor(HIF)in placenta of pregnant women with intrahepatic cholestasis of pregnancy(ICP).
HIF-1α and HIF-2α Proteins from villous explant culture under hypoxia condition were probed by immuno-histochemistry methods.
① HIF-1α protein in the placental villous tissues cultured for 4 h under hypoxic condition(2% O2)were detected in two groups.The expressions of HIF-1α increased in the placentae of pregnant women with ICP(P<0.05).With the O2 concentration recovered, the high O2 concentration(21% O2)decreased the expression HIF-1α in 15 min(P<0.05)in two groups.②The expression of HIF-2α was highly increased under hypoxia condition than that under normal concentration O2(P<0.01); 21 % O2 decreased the expression HIF-2α in 15 min(P<0.01)in normal group.But no significant difference was revealed in the change of the HIF-2α in ICP(P > 0.05).
Our research suggested that as a sensitive index, high increase of HIF-la may play an important pole in adapting hypoxia and the abnormal change of HIF-2α may also bring bad pregnancy outcome in ICP under hypoxic conditions.
