Study on the expression of mRNA for MCP-1 in cerebral tissue of newborn rats with experimental hypoxic-ischemic brain damage

Objective

To observe the expression of mRNA for the MCP-1 in cerebral tissue after experimental hypoxic-ischemic brain damage(HIBD)in newborn rats.

Methods

The HIBD model of SD rat was set up by ligating the right carotid artery and exposing the animals to 8% oxygen for 2. 5 hours. The dynamic changes of expression of MCP-1 mRNA were studied by using quantitative, real-time, fluorogenic PCR(TaqMan)assay.

Results

Peaking expression of MCP-1 mRNA was shown at 6 hours after HIBD and the level of MCP-1 mRNA in the hypoxic-ischemic(HI)cerebral hemisphere was significantly higher than that of the control group(P<0. 05). The level of MCP-1 mRNA in the HI cerebral hemisphere remained greater than that of the control group even at 12 hours after HIBD(P< 0. 05). The expression of MCP-1 mRNA declined at 24〜72h after HIBD in the HI cerebral hemisphere and there was no difference compared with that in the control group(P>0. 05).

Conclusion

It is suggested that as inflammatory mediators, the MCP-1 expression after HIBD may play an important role in the mechanism of brain damage.
