目的

通过对临床子宫内膜异位症(endometriosis, EMS)患者异位内膜及在位内膜survivin mRNA,bcl-2 mRNA及cox-2 mRNA表达的测定,探讨SurvivinBcl-2及Cox-2在子宫内膜异位症发病及发展中的分子生物学机制及其三者的相关性。

方法

选取2006年1月至2006年11月,在新疆医科大学第一附属医院妇科接受开腹手术或腹腔镜手术治疗的子宫内膜异位症患者32例为研究对象(EMS组),并在术中取得异位内膜组织、在位内膜组织,另取正常妇女(除外激素依赖性疾病或激素治疗患者)正常子宫内膜32例(对照组)(分组征得患者本人的知情同意),应用半定量逆转录-聚合酶链反应(RT-PCR)方法,测定survivin,bcl-2及cox-2的mRNA表达水平。

结果

与对照组相比,EMS组患者异位内膜的survivin mRNA,bcl-2 mRNA的表达水平上调,cox-2 mRNA的表达水平也明显上调,且总体survivin,bcl-2及cox-2的mRNA表达与子宫内膜异位症呈正相关,三者有明显相关性。而EMS组在位内膜与之相比,Survivin上调,Bcl-2,Cox-2变化差异无显著意义。

结论

子宫内膜异位症患者SurvivinBcl-2及Cox-2上调,可能是子宫内膜异位症发生发展的分子生物学机制之一,与异位内膜的发生和种植有关,且三类基因在疾病的发展中关系密切。

Objective

To evaluate the molecular mechanism by measuring the gene expression of Survivin, Bcl-2 and Cox-2 in patients with endometriosis(EMS) and investigate their effects on the initiation and maintenance of endometriosis.

Methods

From January 2006 to November 2006, the ectopic endometrium and eutopic endometrium were obtained from 32 patients with endometriosis (EMS group) and from 32 normal endometrium(the control group). Informed consent was obtained from all patients. The mRNA expression of survivin, bcl-2 and cox-2 were measured by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR).

Results

Compared to normal group, the mRNA levels of survivin of ectopic endometrium and eutopic endometrium were significantly increased, the mRNA levels of bcl-2, cox-2 were significantly increased too. The total mRNA levels of survivin, bcl-2 and cox-2 were positively correlated with endometriosis, and the correlations of three were obvious.

Conclusion

Abnormal up-regulation of Survivin, Bcl-2 and Cox-2 in the endometriosis patients might be one of molecular biologic mechanisms for ecotopic implantation and development of endometrium. Furthermore, three genes were highly associated to the endometriosis progress.
