目的

探讨华蟾素对体外培养的卵巢癌3AO细胞芳香化酶和雌二醇表达的影响和意义。

方法

体外培养卵巢癌3AO细胞,不同浓度的华蟾素干预后,应用RT-PCR检测芳香化酶的表达,放射免疫法检测雌二醇的表达,流式细胞仪检测细胞的凋亡。

结果

卵巢癌3AO细胞对照组的芳香化酶mRNA表达为1.39±0.13,其合成的雌二醇为(8.69±13.46)pmol/L,细胞凋亡率为2.31±0.98% ;0.25 mg/ml华蟾素对卵巢癌3AO细胞芳香化酶mRNA和雌二醇的表达以及凋亡率无影响(P>0.05);2.5 mg/ml华蟾素干预后,卵巢癌3AO细胞芳香化酶mRNA表达为0.87±0.16,雌二醇为(10.42±4.23)pmol/L,癌细胞凋亡率为28.69±4.16%,均与卵巢癌3AO细胞对照组有统计学差异(P<0.05);25 mg/ml华蟾素与2.5 mg/ml华蟾素对卵巢癌3AO细胞芳香化酶mRNA、雌二醇表达和癌细胞凋亡率的影响无统计学差异(P>0.05)。

结论

卵巢癌3AO细胞可利用芳香化酶合成内源性雌二醇,2.5 mg/ml华蟾素即可抑制癌细胞芳香化酶mRNA的表达和雌二醇的合成,抑制癌细胞的增殖。

Objective

To investigate the effect and significance of cinobufacine on the expression of aromatase and estrodiol in the ovarian carcinoma 3AO cells cultured in vitro.

Methods

Ovarian carcinoma 3AO cells were cultured in vitro ; after the action of cinobufacine with different concentration, the expression of aromatse was measured by RT-PCR ; the expression of estrodiol was determined by radioimmunoassay, the cellular apoptosis was tested by flow cytometry.

Results

Ovarian carcinoma 3AO cells expressed aromatase mRNA of 1.39 ± 0.13,the estrodiol was(8.69 ± 13.46)pmol/L,the cellular apoptotic rate was 2.31 ± 0.98% ; 0.25 mg/ml cinobufacine didn't affected the aromatase 、estradiol and cellular apoptotic rate(P>0.05); after the action of 2.5 mg/ml cinobufacine, the expression of aromatase was 0.87 ± 0.16, the concentration of estradiol was(10.42 ± 4.23)pmol/L , the cellular apoptotic rate was 28.69 ± 4.16%, all of these deta were different from the control group of ovarian carcinoma 3AO cells significantly(P < 0.05); the action of 25 mg/ml cinobufacine was similar to that of 0.25 mg/ml cinobufacine(P>0.05).

Conclusion

Ovarian carcinoma 3AO cells can synthesize endogeneous estradiol by aromatase, 2.5 mg/ml cinobufacine inhibit the espression of aromatase mRNA and the synthesis of estradiol, and inhibit the proliferation of the ovarian carcinoma cells.
