目的

探讨妊娠对SD大鼠的体表痛阈值及七氟醚最低肺泡有效浓度(MAC)的影响。

方法

选择同一批次的健康(清洁级)雌性SD大鼠40只为研究对象,按照是否妊娠及妊娠不同时期,将其分为4组,早孕期组(n=10,妊娠6 d)、中孕期组(n=10,妊娠12 d) 、晚孕期组(n=10,妊娠18 d)及对照组(n=10,未孕大鼠)(本研究所遵循的程序符合实验动物保护与使用准则)。采用辐射热甩尾法测定大鼠痛觉,用秒表测定大鼠甩尾所需反应时间(s)并作为体表痛阈值。应用夹尾刺激法测量4组大鼠七氟醚MAC值。

结果

①4组SD大鼠的体质量比较,差异有统计学意义(F=41.394,P=0.000)。②4组SD大鼠的体表痛阈值比较,差异有统计学意义(F=3.531,P=0.007)。③4组SD大鼠的七氟醚MAC值比较,差异有统计学意义(F=35.889,P=0.000),其中,早孕期组和中孕期组比较,差异无统计学意义(P=0.659),其余各组两两比较,差异均有统计学意义(P<0.001)。

结论

妊娠期大鼠对外界刺激的神经灵敏度和麻醉药物用药量增加。

Objective

To investigate effects of somatic pain threshold and sevoflurane minimum alveolar concentration (MAC) in rats during pregnancy.

Methods

A total of 40 Sprague-Dawley female rats (supplied by the animal center of West China School of Medicine, Sichuan University) were recruited into this study. The rats were divided into 4 groups, first trimester group (n=10, 6 days old pregnancy), second trimester group (n=10, 10 days old pregnancy), third trimester group (n=10, 18 days old pregnancy) and control group (n=10, unpregnancy). Tail flick test was used to test rat algesthesia, and stopwatch was used to record the time (s) for tail flick that was taken as the value of somatic pain threshold. Sevoflurane MAC determination was performed by clipping rat tail.

Results

①The statistically significant difference was found in weight among 4 groups (F=41.394, P=0.000). ② There was significant difference in somatic pain threshold among 4 groups(F=3.531, P=0.007). ③ There was significant difference in sevoflurane MAC values among 4 groups(F=35.889, P=0.000), and the statistically significant difference was not found between first trimester group and second trimester group (P=0.659), but there was significant difference among the rest groups(P<0.05).

Conclusions

The nerves system sensitivity and dose of narcotic drugs increased obviously in rats during pregnancy.
