目的

探讨新生SD大鼠缺氧缺血性脑损伤(HIE)后,应用金纳多及姜黄素治疗后其脑组织中基质金属蛋白酶(MMP)-9的表达变化趋势。

方法

将新生7 d SD大鼠90只随机分为5组,假手术组、HIE组、HIE+金纳多干预组(金纳多治疗组)、HIE+姜黄素干预组(姜黄素治疗组)及HIE+金纳多+姜黄素干预组,每组各18只。HIE模型制备成功后处死各动物,观察HIE后6 h,24 h和48 h,观察脑组织MMP的表达。

结果

HIE模型制备成功后,各组SD大鼠行为出现不同改变。金纳多治疗组、姜黄素治疗组和综合治疗组于各时间亚组MMP-9免疫阳性细胞平均灰度值分别与假手术组、HIE组比较,差异均有统计学意义(P<0.01);金纳多治疗组、姜黄素治疗组各时间亚组分别与综合治疗组比较,差异亦有统计学意义(P<0.01)。

结论

金纳多+姜黄素对新生SD大鼠HIE后神经系统功能恢复具有更为明显的改善作用。

Objective

To explore the matrix metalloproteinase (MMP)-9 expression changes of newborn rats with hypoxic-ischemic encephalopathy (HIE) after ginaton and curcumin treatment.

Methods

Ninety newborn 7 days Sprague-Dawley rats were randomly divided into five groups, sham operation control group (n=18), HIE group(n=18), HIE+ ginaton intervention group (ginaton group, n=18), HIE+ curcumin intervention group(curcumin group, n=18), HIE+ ginaton+ curcumin intervention group(comprehensive group, n=18). Those five groups were further divided into three sub-group after HIE, 6 h sub-group, 24 h sub-group, 48 h sub-group. The change tendency of MMP-9 after treating by ginaton and curcumin were observed.

Results

Compared with sham group and HIE group, the 6 h, 24 h, and 48 h sub-group of immunoreactive cells in the average gray value among ginaton group, curcumin group comprehensive group had statistically significant difference (P<0.01), and there still had statistically significant difference between ginaton group and comprehensive group, curcumin group and comprehensive group (P<0.01).

Conclusion

The effects of ginaton combined with curcumin had obvious improvement on the nervous system functions recovery after HIE.
